DNA repair is a critical process to correct DNA damage. If the process fails, irreparable DNA damage may occur and lead to malignant tumors or cancers.
 
Both environmental factors and cellular metabolic processes may cause DNA damage. Replication error is one source of DNA damage. Most of the error replicated bases are removed by the proofreading activity of DNA polymerase. The missed error bases are later corrected by mismatch repair (MMR) system which scans newly replicated DNA to identify and excise the mismatched base specifically from the newly replicated DNA strand, allowing the error to be corrected.
 
MMR proteins work coordinately in sequential steps to initiate repair of DNA mismatches. Mutations in MMR proteins result to microsatellite instability (MSI), a condition of genetic hypermutability, and implicate in many cancers.
