GDN and LMNG Detergents Now Available

Ideal For Cryo-EM and X-Ray Crystallography Workflows

GDN and LMNG are two of the most widely used detergents in the structural biology workflow used in tandem for solubilisation and purification of membrane proteins. Both show success in Cryo-EM and X-ray crystallography.

Glyco-diosgenin (GDN)

Glyco-diosgenin (GDN) is a popular synthetic drop-in substitute for Digitonin. As a synthetic compound, there is no batch-to-batch variability and no toxic byproducts.
GDN is compatible with the lipidic cubic phase (LCP) crystallisation method, where the cubic phase formed by the host lipid monoolein is not disrupted by concentrations of GDN up to 20 mM (2.3%) and over a wide range of temperatures.
Typical concentrations of GDN used for protein purification and Cryo-EM structure determination range between 0.17 mM – 0.5 mM (0.02% - 0.06%); well below the concentration where the cubic phase is destabilised.
Membrane proteins that are stable and functional in GDN can proceed directly into LCP crystallisation trials.

Lauryl Maltose Neopentyl Glycol (LMNG)

Lauryl Maltose Neopentyl Glycol (LMNG) is an amphiphilic detergent ideally suited to improve solubilisation of integral membrane proteins while maintaining structural integrity and activity.
In recent years, the use of LMNG mixed with cholesteryl hemisuccinate (CHS) has gained in popularity for the solubilisation, purification, and structure determination of membrane proteins. LMNG is typically used at a ten-fold excess over CHS.
The most used concentration for membrane protein solubilisation is 1% LMNG : 0.1% CHS. For membrane protein purification and structure determination, the commonly used concentration is 0.01% LMNG : 0.001% CHS.
In 2023, GDN has been used in combination with LMNG and CHS to solve the Cryo-EM structure of the pivotal role of arresting regulating GPCRs. (PDB IDs: 7R0C, 6UP7, and 7SRS).
